Ruthenium red staining of polyanion containing structures in sections from epoxy-resin embedded tissues

Summary Staining by ruthenium red (0.5 mg/ml in borate buffer at pH = 9.2) has been used for light and electron microscopic visualization of polyanion containing structures in sections from glutaraldehyde-fixed, epoxy-embedded tissues. This staining technique can be applied in a simple and rapid way, showing the reactive cell components with suitable resolution and contrast. Preliminary spectrophotometric studies show the correspondence in absorption characteristics of the dye which is bound to polyanions in situ or in vitro .

Rainfall drop size distribution in the Western Mediterranean basin, València, Spain

Although soil erosion processes are directly related to the raindrop sizes and the kinetic energy of the rainfall, little is known about the drop-size distribution. The paper staining technique is used to measure the size of the raindrops for natural thunderstorms with different rainfall intensities in the Western Mediterranean basin (Canals and Valencia, Spain). Drop sizes are greater during the most intense thunderstorms, ranging from 0.25 mm diameter (D50) for a 1 mm h−1 rainshower to 2.69 mm in very intense thunderstorms of about 120 mm h−1. When collection of ejaculated sperm samples is not possible, as is the case with wild species, the epididymides of sacrificed wild males become the only possible source of spermatozoa. Mature cauda epididymal spermatozoa display characteristics similar to those of ejaculated sperm cells. The present work proposes a sperm staining technique suitable for the morphometric evaluation of red deer epididymal sperm using a new computerized system.
